Scientific Report IL-23 drives differentiation of peripheral cd17 T cells from adult bone marrow-derived precursors

چکیده
Pro-inflammatory interleukin (IL)-17-producing cd (cd17) T cells are thought to develop exclusively in the thymus during fetal/perinatal life, as adult bone marrow precursors fail to generate cd17 T cells under homeostatic conditions. Here, we employ a model of experimental autoimmune encephalomyelitis (EAE) in which hematopoiesis is reset by bone marrow transplantation and demonstrate unequivocally that Vc4 cd17 T cells can develop de novo in draining lymph nodes in response to innate stimuli. In vitro, cd T cells from IL-17 fate-mapping reporter mice that had never activated the Il17 locus acquire IL-17 expression upon stimulation with IL-1b and IL-23. Furthermore, IL-23R (but not IL-1R1) deficiency severely compromises the induction of cd17 T cells in EAE, demonstrating the key role of IL-23 in the process. Finally, we show, in a composite model involving transfers of both adult bone marrow and neonatal thymocytes, that induced cd17 T cells make up a substantial fraction of the total IL-17-producing Vc4 T-cell pool upon inflammation, which attests the relevance of this novel pathway of peripheral cd17 T-cell differentiation.

Objective(s): Cells perform their functional activities by communicating with each other through endogenous substances and receptors. Post-translation, stem cells function properly in new host tissue by carrying specific cell surface receptors. We aimed to characterize muscarinic receptor subtypes in mesenchymal stem cells (MSCs) together with osteogenic and adipogenic...
متن کاملSertoli cell condition medium can induce germ like cells from bone marrow derived mesenchymal stem cells
Objective(s): Although many researchers have confirmed induction of germ cells from bone marrow mesenchymal stem cells (BMMSCs), there are no reports that confirm spontaneous differentiation of germ cells from BMMSCs. In this study, we have evaluated the effect of adult Sertoli cell condition medium (SCCM) as a mutative factor in the induction of germ cells from BMMSCs. Materials and Methods: ...
متن کاملCFU-GM Like Colonies Derived from Embryonic Stem Cells Cultured on the Bone Marrow Stromal Cells
The aim of this study was to isolate mouse embryonic stem cells from late blastocyst stage embryos and to use them as a model system for the study of hematopoietic induction outside the embryo by coculturing of embryonic stem cells with bone marrow stromal cells. Blastocyst stage embryos from pregnant NMRI mice were obtained and cultured for 1-2 days in DMEM medium. The inner cell masses formed...

Trying to use embryonic stem cells about 20 years ago, working with animals, especially rats began. During these years , many experiments in mouse embryonic stem cells to transform into a variety of cells and transplanting them were led to remarkable success . In the next issue of human stem cells were considered successful until finally in 1998 the first report was published in the proliferat...
